The purpose is to use PCR to amplify a gene from the influenza virus (the matrix gene). I want to find the optimal concentration of MgCl2 (one of the reagents) for a reverse-transcription real-time PCR that would produce results exhibiting efficiently produced products. Efficiency is analyzed in terms of:
  • The amount product is formed in a given time (the reaction requiring a minimal amount of time for initial production)
  • Comparing the growth of PCR product to the theoretically expected exponential growth of product
