After thirty minutes incubation, almost 100% of the simvastatin and 30% of the pravastatin was metabolized when no other substrates were added. When itraconazole was added, the amount of simvastatin metabolized dropped to only about 30%. Itraconazole is a known inhibitor of CYP3A4, and its effect on the simvastatin proves that simvastatin is metabolized by CYP3A4.
